Structural analyses of PCNA from the fungal pathogen Candida albicans identify three regions with species‐specific conformations

چکیده

An assembly of multiprotein complexes achieves chromosomal DNA replication at the fork. In eukaryotes, proliferating cell nuclear antigen (PCNA) plays a vital role in fork and is essential for viability. PCNA from several organisms, including Saccharomyces cerevisiae , has been structurally characterised. However, structural analyses fungal pathogens are limited. Recently, we have reported that opportunistic pathogen Candida albicans complements functions ScPCNA S. . Still, it only partially rescues loss when yeast cells under genotoxic stress. To understand this further, herein, determined crystal structure CaPCNA compared with existing structures other human PCNA. Our comparative in‐solution small‐angle X‐ray scattering (SAXS) reveal forms stable homotrimer, both solution. It displays noticeable alterations oligomerisation interface, P‐loop hydrophobic pocket regions, suggesting its differential function heterologous system avenues developing specific therapeutics. Databases The PDB SASBDB accession codes 7BUP SASDHQ9, respectively.

Regulation of phenotypic transitions in the fungal pathogen Candida albicans

The human commensal fungus Candida albicans can cause not only superficial infections, but also life-threatening disease in immunocompromised individuals. C. albicans can grow in several morphological forms. The ability to switch between different phenotypic forms has been thought to contribute to its virulence. The yeast-filamentous growth transition and white-opaque switching represent two ty...

Chemogenomic Profiling of the Fungal Pathogen Candida albicans

There is currently a small number of classes of antifungal drugs, and these drugs are known to target a very limited set of cellular functions. We derived a set of approximately 900 nonessential, transactivator-defective disruption strains from the tetracycline-regulated GRACE collection of strains of the fungal pathogen Candida albicans This strain set was screened against classic antifungal d...

Glucanase Induces Filamentation of the Fungal Pathogen Candida albicans

Candida albicans is the most common human fungal pathogen. Many organisms, including C. albicans, secrete glucanases under different environmental conditions. Here, we report a novel role for beta-1, 3- glucanase in inducing Candida albicans to form filaments at 22°C and enhancing filamentation at 37°C in nutrient-rich medium. Quorum sensing, the efg1-signaling and cek1 MAP kinase pathways are ...

Oxidative Stress Responses in the Human Fungal Pathogen, Candida albicans

Candida albicans is a major fungal pathogen of humans, causing approximately 400,000 life-threatening systemic infections world-wide each year in severely immunocompromised patients. An important fungicidal mechanism employed by innate immune cells involves the generation of toxic reactive oxygen species (ROS), such as superoxide and hydrogen peroxide. Consequently, there is much interest in th...

Glucose promotes stress resistance in the fungal pathogen Candida albicans.

Metabolic adaptation, and in particular the modulation of carbon assimilatory pathways during disease progression, is thought to contribute to the pathogenicity of Candida albicans. Therefore, we have examined the global impact of glucose upon the C. albicans transcriptome, testing the sensitivity of this pathogen to wide-ranging glucose levels (0.01, 0.1, and 1.0%). We show that, like Saccharo...
